Gene therapy is the insertion of genes into an individual's cells and tissues to treat a disease, and hereditary diseases in which a defective mutant allele is replaced with a functional one. Although the technology is still in its infancy, it has been used with some success. Antisense therapy is not strictly a form of gene therapy, but is a genetically-mediated therapy and is often considered together with other methods.

La thérapie génique est une stratégie thérapeutique qui consiste à faire pénétrer des gènes dans les cellules ou les tissus d'un individu pour traiter une maladie. Mit Gentherapie bezeichnet man das Einfügen von Genen in Zellen eines Individuums zur Behandlung von Erbkrankheiten bzw. Gendefekten. Durch die Einführung dieser Gene kann ein genetischer Defekt kompensiert werden. Ein genetischer Defekt liegt vor, wenn bei einem Lebewesen ein Gen fehlt oder eine Mutation enthält, die dazu führt, dass das Genprodukt nicht gebildet wird oder seine Funktion nicht richtig ausüben kann.

Per terapia genica (a cui ci si riferisce comunemente anche con il termine inglese di Gene Therapy) si intende l'inserzione di materiale genetico (DNA) all'interno delle cellule al fine di poter curare delle patologie. Questa procedura di inserzione è nota come transfezione.La sua concezione si sviluppò a seguito del grande progresso delle metodiche di biologia molecolare sviluppatesi dagli anni '80. Tali tecniche consentirono il clonaggio ed il sequenziamento di vari geni. Ciò comportò la precisa identificazione di molte alterazioni geniche in diverse patologie e la capacità, grazie alle tecniche del DNA ricombinante, di modificare microorganismi (come batteri o funghi) per poter far loro esprimere delle molecole d'interesse.
