epitope

An epitope is the part of a macromolecule that is recognized by the immune system, specifically by antibodies, B cells, or T cells. Although epitopes are usually thought to be derived from nonself proteins, sequences derived from the host that can be recognized are also classified as epitopes.Most epitopes recognized by antibodies or B cells can be thought of as three-dimensional surface features of an antigen molecule; these features fit precisely and thus bind to antibodies. The part of an antibody that recognizes the epitope is called a paratope. Exceptions are linear epitopes, which are determined by the amino acid sequence (the primary structure) rather than by the 3D shape (tertiary structure) of a protein.

Un épitope, aussi appelé déterminant antigénique, est une molécule qui peut être reconnue par un paratope (partie variable d'un anticorps ou d'un récepteur membranaire des lymphocytes T : TCR), pour déterminer si elle appartient au domaine du soi ou au domaine du non-soi. Un antigène est caractérisé par ses épitopes, si ses épitopes sont reconnus comme appartenant au non-soi alors il est lui-même immédiatement reconnu comme appartenant au non-soi. Cette reconnaissance épitope/paratope est donc à la base de la réponse immunitaire spécifique : elle permet la sélection clonale, c’est-à-dire la sélection des acteurs capables de s'attaquer spécifiquement à l'antigène correspondant à un épitope particulier.

Ein Epitop (auch antigene Determinante genannt) ist ein kleiner Bereich (Molekülabschnitt) eines Antigens, gegen den das Immunsystem Antikörper bildet; denn weder die Rezeptoren auf der Oberfläche von B-Zellen noch die Antikörper selbst können an ein ganzes Antigen binden. Antigene Determinanten sind in der Natur Proteine bzw. Proteide. Es können aber auch Antikörper gegen andere chemische Strukturen (z.B. Hormone) gebildet werden. Diese Antikörper werden u.a. in der Immunanalytik verwendet.

Epitopem lub determinantą antygenową nazywamy fragment antygenu, który łączy się bezpośrednio z przeciwciałem. Jeżeli np. mamy bakterię gramujemną (antygen), to przeciwciała mogą wiązać się z jej rzęskami, białkami powierzchniowymi i lipopolisacharyd (epitopy na powierzchni antygenu). Z kolei każdy z tych epitopów także może być potraktowany jako antygen, a wtedy np. określone miejsce rzęski, do którego przyłącza się przeciwciało, też można potraktować jako epitop. Tak więc pojęcie epioptu zależy od skali. Najbradziej ścisłe określenie epitopu będzie oznaczać tylko i wyłącznie miejsce bezpośredniej interakcji antygenu z przeciwciałem (a konkretnie z paratopem).
