Disulfiram is a drug used to support the treatment of chronic alcoholism by producing an acute sensitivity to alcohol. Trade names for disulfiram in different countries are Antabuse and Antabus manufactured by Odyssey Pharmaceuticals. Disulfiram is also being studied as a treatment for cocaine dependence, as it prevents the breakdown of dopamine (a neurotransmitter whose release is stimulated by cocaine); the excess dopamine results in increased anxiety, higher blood pressure, restlessness and other unpleasant symptoms. Several studies have reported that it has anti-protozoal activity as well.

Disulfiram (Tetraethylthiuramdisulfid) ist ein Alkoholentwöhnungsmittel, das bei Alkoholkranken angewendet werden kann.Sobald Alkohol in geringen Dosen eingenommen wird, entstehen starke und unangenehme Unverträglichkeitsreaktionen wie Hautrötung, Kopfschmerzen, Kältegefühl in den Extremitäten und Übelkeit, oft auch Herz-Kreislauf-Beschwerden (Acetaldehydsyndrom). Diese Wirkung, das Acetaldehydsyndrom, beruht auf der Blockierung des Enzyms Aldehyddehydrogenase (ALDH). Die Metabolisierung von Alkohol durch die Alkoholdehydrogenase (ADH) bleibt auf der Stufe von Acetaldehyd stehen, es kommt also zu einer Aldehydvergiftung.

Disulfiram - lek stosowany w leczeniu alkoholizmu, którego działanie polega na wpływie na metabolizm alkoholu etylowego. Związek ten zaburza metabolizm alkoholu w organizmie. Hamuje działanie enzymu - dehydrogenazy aldehydowej, przez co utrudnia utlenienie aldehydu octowego (trujący związek) do mniej szkodliwego kwasu octowego. Kumulujący się w organizmie aldehyd octowy powoduje objawy zatrucia (tzw. reakcja disulfiramowa).

Disulfiram is een medicament dat wordt gebruikt bij de behandeling van (chronisch) alcoholisme. Het zorgt er voor dat de patiënt bij alcoholinname geconfronteerd wordt met onaangename effecten zoals onwel worden, zich doodziek voelen, braken, hoofdpijn, versnelde hartslag, transpiratie, rood aanlopen, etc. De werking van het middel is de angst voor de onaangename effecten die het inboezemt bij de ex-verslaafde wanneer hij/ zij alcohol inneemt. Disulfiram wordt onder andere op de markt gebracht onder de naam Refusal en Antabus.
