allosteric regulation

In biochemistry, allosteric regulation is the regulation of an enzyme or protein by binding an effector molecule at the protein's allosteric site (that is, a site other than the protein's active site). Effectors that enhance the protein's activity are referred to as allosteric activators, whereas those that decrease the protein's activity are called allosteric inhibitors. The term allostery comes from the Greek allos, "other," and stereos, "space," referring to the regulatory site of an allosteric protein's being separate from its active site. Allosteric regulation is a natural example of feedback control.

L'allostérie (du grec ἄλλως, allos : autre et στερεός, stereós : forme) est un mode de régulation de l'activité d'une enzyme par lequel la fixation d'une molécule effectrice en un site modifie les conditions de fixation d'une autre molécule, en un autre site distant de la protéine. Ce concept a été formalisé par Jacques Monod, Jean-Pierre Changeux et Jeffries Wyman dans une série d'articles, dont le plus important a été publié en 1965 dans Journal of Molecular Biology.

Der Begriff Allosterie - von griech. allo (anders) und steros (Ort), das heißt " am anderen Ort" - entstammt dem Bereich der Biochemie, dort der Proteinfunktion. Er wird in der Literatur unterschiedlich gebraucht. So gilt er zunächst für Proteine, die bestimmte regulatorisch wirksame Moleküle (Effektoren) an anderer Stelle binden als ihr Substrat (Enzym) bzw. ihren zentralen Liganden (Carrier bzw. Rezeptor). Fälle, in denen die Bindungsstärke von der Zahl der bereits gebundenen Substrat- oder Ligandenmoleküle abhängt, gehören ebenfalls dazu.
