Tetanus toxin is an extremely potent neurotoxin produced by the vegetative cell of Clostridium tetani in anaerobic conditions, causing tetanus. It has no known function for clostridia in the soil environment where they are normally encountered. It is also called spasmogenic toxin, tetanospasmin or abbreviated to TeTx or TeNT. The LD₅₀ of this toxin has been measured to be approximately 1 ng/kg, making it second only to Botulinum toxin D as the deadliest toxin in the world. However, these tests are conducted solely on mice which may react differently to certain toxins than humans and other animals.

Das Tetanustoxin oder Tetanospasmin ist das wichtigste der von Clostridium tetani gebildeten Exotoxine. Es wirkt direkt neurotoxisch. Das Tetanospasmin ist ein 150 kDa großes Protein. Es wird teils aktiv von den Bakterien sezerniert, teils wird es bei Lyse von Bakterien freigesetzt. Das Toxin besteht aus 2 Untereinheiten. Die schwere Protein-Kette ist verantwortlich für die Bindung des Toxins an Ganglioside von Nervenzellen, während die leichte Kette die Neurotoxizität vermittelt. Das Toxin hemmt präsynaptisch die inhibitorischen Synapsen an den spinalen Motoneuronen sowie die Freisetzung der Neurotransmitter Glycin und GABA. Klinisch zeigt sich dieses in einer spastischen Paralyse. Tetanospasmin scheint in der Regel Plasmid-codiert zu sein.
