The endomembrane system is composed of the different membranes that are suspended in the cytoplasm within a eukaryotic cell. These membranes divide the cell into functional and structural compartments, or organelles. In eukaryotes the organelles of the endomembrane system include: the nuclear envelope, the endoplasmic reticulum, the Golgi apparatus, lysosomes, vacuoles, vesicles, peroxisomes and the cell membrane. The system is defined more accurately as the set of membranes that form a single functional and developmental unit, either being connected together directly, or exchanging material through vesicle transport. Importantly, the endomembrane system does not include the membranes of mitochondria and chloroplasts.

Als Endomembransystem wird die Gesamtheit der membranumschlossenen Zellkompartimente bezeichnet, die durch Vesikeltransport miteinander verbunden sind. Es setzt sich aus dem Endoplasmatischen Reticulum (raues und glattes), dem Golgi-Apparat, der Plasmamembran, der äußeren Kernhülle bzw. dem perinucleären Raum, Endosomen, Lysosomen und Transportvesikeln zusammen; im Gegensatz dazu sind Mitochondrien, Plastiden und Peroxysomen kein Bestandteil des Endomembransystems. Das Lumen all dieser Organellen wird als exoplasmatisch bezeichnet, d.h. alles, was von diesen Membranen eingeschlossen ist, kann aus der Zelle herauskommen, ohne eine Membran zu überschreiten.
